Topic: Reverse Vaccinology Approach to Design a Novel Multi-Epitope Vaccine Candidate against COVID-19: An In Silico Study
Abstract
A new multi-epitope vaccination targeting 2019-nCoV that can elicit CD4+ and CD8+ T-cell immunological responses was produced in this study. The virus's three known antigenic proteins were chosen, and the possibility of immunogenic B and T-cell epitopes was examined. The durability of the fusion protein containing the receptor TLR4 and HLA-A was assessed using MD simulation. In this work, bioinformatics methods were used to assess ORF10, ORF3A, ORF10, N, E and M proteins in order to form a multi-epitope vaccine for the activation of immunological responses for coronavirus.
Introduction
The coronavirus disease COVID-19 epidemic began in Wuhan, the capital of Hubei province in late December 2019. All world the world, scientists want to make a vaccine to control this disease. In this article, scientists want to design a multi-epitope vaccine candidate against coronavirus. In our research, scientists want to discover multi-epitope vaccine candidates using innate immunity receptors (Toll-like receptors TRL4) and cellular immunity receptors (HLA-A).
Literature Review
The covid-19 contains many structural many non-structural proteins. proteins. Proteins like S protein (spike), E protein (envelope), M protein (membrane), and N protein (nucleocapsid) are structural proteins. Because of its antigenicity and highly conserved structure, the N protein is a good candidate for vaccination. It can also induce strong antibody production and it can trigger cytokine reaction. M protein is also a suitable target because it has a role in virions and with the help of S protein it can make interaction with the genomic RNA.
Methodology
At first, we will select a better vaccine candidate against coronavirus. To choose a suitable vaccine Candidate, amino acid structures of protein which are found in covid 19 are retrieved in FASTA Sequence from NCBI. We have selected six proteins. Their antigenic analysis, peptide binding to MHC molecules, epitope-rich domains, physiochemical parameters, secondary and tertiary structures, protein-protein molecular docking, molecular dynamics and binding energy were evaluated by bioinformatics servers on NCBI.
Results and Discussion
The conclusion is that for the future candidate vaccine, NOM protein will consider against Covid-19 because it can trigger both humoral immunity and cellular immunity. In about 100ns, the NOM-TRL4 and NOM-HLA complexes were extremely stable, with strong molecular Interactions. The whole vaccination candidates were studied using molecular simulation, Molecular docking, and structural analysis. The concept of a safe and effective vaccination might help to limit the number of people who suffer from this disease. In vaccine development, Immunoinformatics technologies are important to save time and money.
